Engineering Nitrogenases for Synthetic Nitrogen Fixation: From Pathway Engineering to Directed Evolution

چکیده
Globally, agriculture depends on industrial nitrogen fertilizer to improve crop growth. Fertilizer production consumes fossil fuels and contributes environmental pollution. A potential solution would be harness nitrogenases—enzymes capable of converting atmospheric N 2 NH 3 in ambient conditions. It is therefore a major goal synthetic biology engineer functional nitrogenases into plants, or bacteria that form symbiotic relationships with crops, support growth reduce dependence industrially produced fertilizer. This review paper highlights recent work toward understanding the requirements for nitrogenase expression manipulating gene heterologous hosts activity oxygen tolerance potentially plants.

Microbial production of natural and non-natural flavonoids: Pathway engineering, directed evolution and systems/synthetic biology.
In this review, we address recent advances made in pathway engineering, directed evolution, and systems/synthetic biology approaches employed in the production and modification of flavonoids from microbial cells. The review is divided into two major parts. In the first, various metabolic engineering and system/synthetic biology approaches used for production of flavonoids and derivatives are di...
متن کاملSynthetic biology approaches to engineering the nitrogen symbiosis in cereals.
Nitrogen is abundant in the earth's atmosphere but, unlike carbon, cannot be directly assimilated by plants. The limitation this places on plant productivity has been circumvented in contemporary agriculture through the production and application of chemical fertilizers. The chemical reduction of nitrogen for this purpose consumes large amounts of energy and the reactive nitrogen released into ...
متن کاملEnzyme engineering: rational redesign versus directed evolution.
Enzyme engineering is undergoing the most profound and exciting transformation in its history (1). It promises unprecedented expansion in the scope and applications of modified or improved enzymes with desired physical and catalytic properties. Two complementary strategies are currently available: rational redesign (2-5) and directed evolution (6,7). Although both approaches have met with great...
متن کاملOptimizing the search algorithm for protein engineering by directed evolution.
An in silico protein model based on the Kauffman NK-landscape, where N is the number of variable positions in a protein and K is the degree of coupling between variable positions, was used to compare alternative search strategies for directed evolution. A simple genetic algorithm (GA) was used to model the performance of a standard DNA shuffling protocol. The search effectiveness of the GA was ...
متن کاملHydroxyapatite from Fish for Bone Tissue Engineering: A Promising Approach
Natural or synthetic hydroxyapatite (HA) has been frequently used as implant materials for orthopaedic and dental applications, showing excellent bioactivity, adequate mechanical rigidity and structure, osteoconductivity and angiogenic properties, no toxicity, and absence of inflammatory or antigenic reactions. HA can be easily synthesized or extracted from natural sources, such as bovine bone....
